PET |
| |
PET is a cell dissociation mix specially formulated for use
with the "Cold Trypsin" method. When used to detach monolayer cultures of
epithelial cells from the growth substratum, this reagent will yield more
viable cells from monoloyers grown in serum-free medium. The solution
contains polyvinylpyrrolidone, EGTA and trypsin in a HBS base. |

Suggested Use: After
rinsing the cell monolayers using HBS, add 3 ml of ice cold PET dissociation
reagent. Do not keep the flask in the incubator. Incubate at room
temperature and monitor the monolayer by microscopy (preferably phase
contrast). When the cells begin to separate, gently remove most of the PET
(without removing the cells) and allow the flask to remain at room
temperature. To determine whether the cells are ready to harvest, tap the
flask (held in an upright position) firmly by hand. When the cells float
free, re-suspend them in cold HBS supplemented with 5% Fetal Bovine Serum to
stop the tryptic action. Mix well by pipetting the cell suspension,
centrifuge and re-suspend the cell pellet in the appropriate complete
serum-free medium. |

Freezing Media Pair |
| |
Freezing Media Pair are used to cryopreserve suspensions of
viable cells detached from monolayer cultures using the "cold trypsin"
method with PET. Cells which have been cryopreserved in this medium have
been shown to have superior viability upon reconstitution. The Freezing
Media Pair has been used extensively to cryopreserve human cells grown in
serum-free media. |

Suggested Use: To
cryopreserve cells, centrifuge the cell suspension. Re-suspend the cells at
4 x 106
cells/ml in ice-cold Freezing Medium A. Slowly add an equal volume of
ice-cold Freezing Medium B to the cell suspension. Mix well. Transfer 1 ml
aliquots of the cell suspension to pre-cooled cryovials. Freeze slowly
using an automatic controlled rate (1°C per minute) freezer to -90°C.
Alternatively, place the cryovials inside a Styrofoam rack and place the
rack in a -80°C mechanical freezer for 24 hours. Transfer the frozen vials
to a liquid nitrogen freezer (vapor phase) for permanent storage. |

FNC COATING MIX® |
| |
FNC COATING MIX®
is formulated to dramatically increase the attachment of cells to the
plastic growth substratum used for culturing mammalian cell lines. Many
fastidious cell types such as human prostate and breast epithelial cells
plated in the appropriate serum-free medium will attach and grow more
effectively on FNC-coated plastic surfaces. |

Suggested Use: To coat
the growth surface, add 0.2 ml FNC COATING MIX®
per square centimeter of surface area of the culture vessel (for example use
5 ml for a T-25 flask). The surface should be completely covered with a
layer of liquid. Incubate for 30 seconds and remove the coating mix with a
pipet. Residual FNC COATING MIX®
will not adversely affect the viability of the cells to be plated.
Do not return any “used” coating mix to the
original container of FNC. This will compromise the performance of the
product. The coating can be done
immediately before seeding the cells or up to one hour beforehand. |

Bovine Pituitary Extract |

Bovine Pituitary Extract prepared from TSE-free animals. The
extracts were prepared from pituitaries harvested from animals in the United
States. The pituitaries were obtained only from USDA inspected facilities
where the animals receive pre- and post-mortem inspection and were found
free of contagious diseases. Foot-and-mouth disease, rinderpest, and
contagious bovine pleuropneumonia are not present in U.S. herds. Further,
there are no known cases of bovine spongiform encephalopathy (BSE) in the
herds from which the pituitaries were obtained. |
