Metrizamide is a tri-iodinated benzamido-derivative of glucose with the
systemic name: 2-
(3-acetamido-5-N-methylacetamido-2,4,6-triiodobenzamido)-2-deoxy-D-glucose.

 | STABILITY AND STORAGE:
When using Metrizamide solutions it is important to ensure that the
conditions used, do not
allow the release of iodine from the aromatic ring, since it is possible
that the released
iodine might modify the biological material being fractionated. Iodine atoms
absorb in the
ultra violet region of the spectrum, and when Metrizamide is irradiated with
ultraviolet
light, the iodine atoms are destabilized. Prolonged exposure to sunlight
and, to a lesser
extent, to diffuse daylight and artifical light can also lead to the release
of iodine from the
molecule. However, the extent of this reaction is generally insignificant
when working
with these solutions on a day to day basis. Iodine is also released when
Metrizamide
solutions are heated above 55°C, so this precludes the use of autoclaving
for sterilizing
solutions. Any iodine released from Metrizamide can be neutralized by the
addition of
either sodium thiosulphate or sodium bisulphate though this does introduce
extra ions into
the solution. |
CTL lymphocytes can be prepared from splenocytes after removal of plastic
adherent cells and centrifugation on a layer of 14.5% (w/v) metrizamide as
described (Macatonia et al (1989) J. Exp. Med. 169, 1255-1264). BAN :
