ISG15 E1 activating enzyme acts on the ubiquitin-like ISG15 protein (also known as UCRP).

The ISG15 (Interferon-Stimulated Gene 15) protein conjugates to target cellular proteins after interferon-alpha/beta stimulation.

The first step of conjugation is activation of ISG15 by ISG15 E1 enzyme: the ATP-dependent formation of a thioester bond between ISG15’s C-terminal carboxyl group and the enzyme.

Next, the activated ISG15 is transferred to the ISG15 conjugating enzyme (Ubc8), and then covalently attached via its C-terminal glycine residue to lysine groups of target proteins—a thiolester conjugation cascade.
