PBS, pH 7.2. |
| Reconstitution |
We recommend a quick
spin followed by reconstitution in 1x PBS, pH 7.2-
7.4 to a concentration of 0.2 mg/mL. This solution can then be diluted into
other aqueous buffers and stored at 4 degree Celsius for 1 week or -20
degree Celsius for future use. |
| Storage |
The lyophilized
protein is stable for a few weeks at room temperature, but
best stored at -20 degree Celsius. Reconstituted IL-12 should be stored in
working aliquots at -20 degree Celsius. |

Activity |
The ED50
as determined by the stimulation of IFN-γ production by mouse
splenocytes co-stimulated with IL-12 is <= 0.1 ng/mL, corresponding to a
specific activity of >= 1 x 10^7 units/mg. |
| Description |
Interleukin-12 is a
regulatory protein produced by activated B-lymphocytes
and macrophages. The biological activities of IL-12 include the stimulation
of growth of activated CD4+ and CD8+T cells and NK cells. Human IL-12 also
promotes the development of proinflammatory/ Th1- like CD4+ cells and
cytotoxic CD8+T cells. Human IL-12 is a disulfide linked heterodimeric
protein with a molecular weight of 70 kDa comprised of disulfide-bonded 35
kDa (p35) and 40 kDa (p40) subunits. |
