An
efficient and economical pre-made screening kit
for establishing the optimal conditions for
the refolding of recombinant protein. |
- We can easily get a target
protein by just expressing it using many kinds of host cells (E.
coli, Yeast, cultured human cells, etc). Among the useful hosts,
E. coli is the most convenient, economical and fastest tool for
protein expression. However, in many cases, over-expression of
foreign proteins results in forming insoluble inclusion bodies.
Regaining the native structure of proteins from inclusion
bodies require screening different conditions including pH,
ionic strength, redox environment, protein concentration,
refolding assistant additive and so on. The task can be
strenuous and take so many times.
Based on our research experience in protein refolding for
many years, BioBud have tried to systemize about 3000 refolding
conditions reported by papers and disclosed patents and
succeeded in determining the most common 384 conditions by
systemically sorting and selecting the various factors affecting
the refolding. Using these 384 conditions, BioBud developed a
pre-made combinatorial screening kit, EasyFolder 3.2 consisting
of four types of unfolding reagents and 96 refolding reagents.
EasyFolder 3.2 allows for the logical evaluation of folding
conditions and getting the optimal condition for refolding of
target proteins.

| - Figure A and B are assay
results from the refolding of serine proteases of similar
structure and activity, using the kit. In the refolding of each
protein, a vast number of conditions such as pl, hydrophobicity
and cystein number, affect the refolding, and a similar result
appears among similar protein. This is a good example of the
necessity of careful selection of the protein refolding
conditions. |
