BACKGROUND:

Endogenous Nitric Oxide (NO) is a diffusible multifunctional second messenger that has been implicated in numerous physiological functions in mammals, such as blood vessel dilation, immune response and synaptic transmission enhancement. The NO is derived from L-arginine as the product of a complex enzymatic reaction catalyzed by a family of three NO synthase (NOS) proteins: nNOS (typeI), iNOS (Type II), eNOS (type III) (1, 2).

The endothelial NOS (eNOS) plays a critical role in controlling vascular tone, platelet aggregation, and cardiac myocyte function (3). It is strongly expressed in malignant mesothelioma, but is not associated with expression of vascular endothelial growth factor (VEGF) and its receptors (4). This phenomenon is different from what has been shown in non-neoplastic endothelial cells.

Rabbit anti-eNOS (Paired116) polyclonal antibody was raised against a synthetic peptide surrounding Serine 116 of bovine eNOS.

REACTIVITY:

This epitope affinity purified rabbit polyclonal antibody is specific for bovine eNOS. The antibodies were evaluated for specificity by immunoblot assay, this antibody recognizes ~130 kDa band in the bovine pulmonary artery endothelial cells lysate, and this immunoreactive band is abolished by the depletion of the immunizing peptide.

APPLICATION:

The working concentrations for specific applications should be determined by the investigator. The results might be affected by several factors, including secondary antibody affinity, antigen concentration, sensitivity of detection method, temperature and length of incubations. ELISA: 0.1-1.0 µg/ml. Western blot: 0.5 to 2 µg/ml. Immunoprecipitation: 3.0-5.0 µg/extract form 107 cells. STORAGE: This polyclonal antibody is supplied as an epitope affinity purified rabbit IgG, 100 µg in 200 µl of phosphate buffered saline (pH 7.4) containing 0.02% sodium azide.
